Products Function
Voriconazole is a broad-spectrum, third-generation synthetic triazole antifungal agent. Its primary mechanism involves inhibiting ergosterol biosynthesis in fungal cell membranes, disrupting membrane integrity and function, thereby causing fungal cell death or growth inhibition.
Products Application
Voriconazole is primarily indicated for treating the following severe invasive fungal infections, particularly common in immunocompromised patients (e.g., organ transplant recipients, chemotherapy-induced neutropenia, HIV/AIDS):
1) . Invasive Aspergillosis: Especially pulmonary aspergillosis, which is its primary indication.
2). Candidiasis: Including candidemia and severe invasive infections caused by fluconazole-resistant Candida species (e.g., Candida krusei).
3) . Other rare but severe fungal infections: caused by Fusarium spp., Scedosporium spp., etc.
4) . Other fungal infections: May also be used to treat cryptococcosis (e.g., cryptococcal meningitis), coccidioidomycosis, histoplasmosis, and penicilliosis.
Mechanism of Action
The mechanism of action of voriconazole is as follows:
1). Inhibition of key enzyme activity: Its active ingredient highly specifically inhibits fungal cytochrome P450-dependent 14α-lanosterol demethylase. This enzyme is a key enzyme in the ergosterol biosynthesis pathway.
2). Disruption of cell membrane synthesis: Inhibition of this enzyme impairs the synthesis of precursors for ergosterol (the primary sterol component of fungal cell membranes) while causing the accumulation of toxic methylated sterol intermediates within fungal cells.
3). Induction of cell death: The absence of ergosterol and accumulation of toxic intermediates severely disrupt the structure and function of fungal cell membranes, increasing membrane permeability and ultimately leading to fungal cell growth inhibition or death.
